Marcus Trescothick made 116 to put Somerset in charge of their rain-affected match at Hove, was Chris Jordan impressed with the ball claiming 5-76.
Jordan removed the second of Somerset's night-watchmen, Alfonso Thomas, early on in the day, trapping the visiting batsmen LBW to earn his third wicket of the innings.
But with Thomas and Craig Overton back in the hutch the scoreboard had a misleading element to it and the partnership between Trescothick and Petersen illustrated the depth remaining in Somerset's batting lineup.
They faced the remainder of the first session with relative ease, with an unbeaten stand worth 85 as Somerset entered lunch with a score of 174-4. Trescothick brought up his fifty in 112 balls and Petersen (76) approached the same landmark, passing fifty just after the interval.
But once their 139-run stand came to an end Somerset's batsmen started to look vulnerable and they lost four wickets for 39 runs, minimising the effort made by Trescothick, who made his 52nd first-class century earlier in the day.
Trescothick's innings was particularly appreciable due to the fact that he made his first ever pair against Sussex last season, with James Anyon dismissing the Somerset opener on both occasions.
Somerset seemed to be heading for an inadequate first innings total, but the final-wicket stand between Johann Myburgh (37*) and Lewis Gregory (47) contained some of the most fluent stroke play all day and added a valuable 75 runs before Somerset were bowled out for 372, with Chris Jordan claiming 5-76.
Sussex's response took an early blow and a seizable one at that, when Ed Joyce, who made consecutive centuries against Warwickshire, edged behind off Alfonso Thomas for six.
Luke Wells (14) and Michael Yardy (5) faced the remainder of play without suffering any further scares with Sussex 25-1 and trailing Somerset by 347 runs.
